This tutorial demonstrates two methods for quantifying dots in a blood plot image: the percent area method and the integrated density method. To use the percent area method, first open the Dot Plot image in ImageJ and invert it to make the dots appear white against a dark background. Select the background noise using the provided selection tools, then click on "Analyze" and measure the intensity, which should be around 54.75. After that, deselect the background noise, go to "Process," select "Math," and subtract the background using the mean intensity value. Ensure the image has a dark foreground and white background by inverting it again. Finally, use the rectangular selection tool to encompass all dots and click on "Analyze," then "Gels," and select "First Lane."
