It involves the application of a small amount of sample directly onto a membrane, which is then probed with a specific antibody or nucleic acid probe to detect the target molecule. Dot Blot may also be used to determine appropriate starting concentration of primary antibody for Western blot.
Dot blot hybridization Dot blot does not require electrophoretic separation of samples. Multiple samples are spotted in same volume next to each other on a single nitrocellulose membrane in dots of equal diameter. Membrane is then hybridized to probe washed and developed similar to northern blotting.
Dot blot is a technique for detecting, analyzing, and identifying proteins, similar to the western blot technique, but differing in that protein samples are not separated electrophoretically but are spotted through circular templates directly onto the membrane or paper substrate.
The main steps in dot blot hybridization are: (1) a small amount of sap is extracted from the plant under test; (2) the viral nucleic acid is denatured by heating or, if it is DNA, by alkali treatment; (3) a spot of the extract is applied to a membrane; (4) the membrane is baked or exposed to ultraviolet light to bind
Interpretation of the results of a dot blot or a slot blot hybridization is relatively straightforward. If hybridization has occurred, a signal is generated in the specific spot. Therefore, a simple yes or no interpretation is usually given. No information is available about the size of the hybridizing fragments.
A direct transfer of RNA or DNA to spots or dots on NC is made. The spotted filter is hybridized with a labeled probe. This method is used on partially degraded RNA samples with good semi-quantitative results. The time required to follow this method is three hours to prepare filter.
Dot blot refers to the deposition of a protein solution directly onto the membrane (20). If the volume to be added to the membrane is small (ca. 5 l), the solution can be applied directly with a capillary micropipette.
