In this tutorial, the presenter demonstrates how to analyze Western blot images using ImageJ. First, open the Western blot image in ImageJ and use the square selection tool to select a band in the first lane. Navigate to the "Analyze" menu and choose "First." To select subsequent bands, use the keyboard arrows to move the square right, repeating the analysis for each lane. Alternatively, the presenter suggests using Ctrl + 1 for the first band and Ctrl + 2 for the others to streamline the selection process. Once all bands are selected, go to "Analyze" and choose "Plot Lanes" to generate graphs. If the resulting graphs display a perfect waveform, use the magic wand tool to finalize the analysis.
