The blot can be laid on the scanner before or after incubation with chemiluminescent substrate. If the substrate hasn't been added, it can be applied directly on the scanner after incubation. Typically, the blot won't dry out while acquiring an image, but a sheet of plastic can be placed on top if extended wetness is needed.
