Avoid These Common Mistakes When Presenting Your Western Blot Images Avoid cosmetic touch-ups. Check your manuscript for placeholders and duplicate figures. Save your original data and consider providing it as public supplement to your readers. Dont mislead. Be careful with splicing and cropping.
Western blots and gels should be submitted as separate files and not embedded in the text. For initial submission, several file types are acceptable, but TIFF or PDF files are preferred. For publication, line art should be 1000 dpi. Color images should be 300 dpi at the final print size.
The main techniques for visualizing a western blot are colorimetric, chemiluminescence, and fluorescence. Colorimetric and chemiluminescence act by an enzymatic reaction either by horseradish peroxidase or alkaline phosphatase (also used in ELISA).
Western Blot Western blotting is a laboratory technique used to detect a specific protein in a blood or tissue sample. The method involves using gel electrophoresis to separate the samples proteins. The separated proteins are transferred out of the gel to the surface of a membrane.
It is very important to be aware that the data produced with a Western blot is typically considered to be semi-quantitative. That is, it provides a relative comparison of protein levels, but not an absolute measure of quantity for a specific target protein in a particular experiment.
Western blots can be detected using: Chemiluminescence and imaged using film or a CCD-based imaging system. Fluorescence and imaged on a fluorescence imaging system. Colorimetric substrates and imaged using white light imaging and a camera.
Chemiluminescent detection methods depend on incubation of the western blot with a substrate that will luminesce when exposed to the reporter on the secondary antibody. The light is then detected by CCD cameras which capture a digital image of the western blot or photographic film.
Open the blot image and the ladder image and convert both images to 8 bit (Image Type 8-bit) Generally invert the blot image so that bands are black and background is white (Edit Invert) Adjust both the blot image and the ladder image (Image Adjust Brightness/Contrast) so that they look good to you.
