Dot blot is a technique for detecting, analyzing, and identifying proteins. This technique is similar to western blot, but protein samples are not separated using electrophoresis; instead, proteins are spotted through circular templates directly onto the membrane or paper substrate.
The western blot is best for detecting a wide range of proteins from a complex mixture or sample. Another key reason to use Western blot is that it is highly specific in measuring proteins, even at relatively low levels.
Dot blot is similar to the other blotting techniques, except that it does not provide information regarding the size of the hybridized fragment. With this technique, extracted DNA or RNA from the target specimen is spotted onto the filter without the prior electrophoresis and transfer steps.
Limited samples: The number of samples assessed in a Western is limited by the number of lanes in the polyacrylamide gel. A dot blot, by comparison, can screen a large number of samples for presence of a POI at once.
A major advantage of dot blot over Western blot is that it is much faster. Dot blot is also more cost-effective, since it eliminates the need for materials such as polyacrylamide gels, molecular weight markers, and all of the equipment and buffers required for gel running and protein transfer.
Limitations of dot-blotting: The dot blot cannot determine the molecular weight of the recombinant protein as the samples are not resolved. It can only show the proteins presence in the cell lysate. However, sometimes it is essential to know whether the protein expressed is of the right size (molecular weight).
The HIV-1 Western blot (WB), the historic gold standard for laboratory diagnosis of HIV-1 infection, is no longer part of the recommended algorithm. The two main reasons for this are the inability of the WB to detect acute infection and the potential to misclassify HIV-2 infection as an HIV-1 infection.
