There are many different immunoassay platforms available to measure to quantitate protein levels in biological fluids. ELISAs are preferred in many cases due to their sensitivity, specificity, accuracy, and ability to tolerate harsh buffers or pretreatments.
The HIV-1 Western blot (WB), the historic gold standard for laboratory diagnosis of HIV-1 infection, is no longer part of the recommended algorithm. The two main reasons for this are the inability of the WB to detect acute infection and the potential to misclassify HIV-2 infection as an HIV-1 infection.
When you need to quantify protein concentration precisely, ELISA is better. Western blot can only measure relative protein abundance and not absolute concentration. The ELISA process is a more straightforward method, too. It uses lower sample volumes and standard 96-well plates and allows for multiplexing.
Generally speaking, an ELISA is easier and faster to perform than a western blot because the protocol is shorter. Moreover, ELISAs are better suited for high throughput labs because they can be performed with lower sample volumes and are usually carried out in 96 well plates, allowing for workflow automation.
RIA is more sensitive but requires special handling and is more expensive, while ELISA is safer, faster, and more cost-effective. RIA is better for detecting low levels of antigens or antibodies in complex biological fluids, while ELISA is better for testing large numbers of samples quickly.
Western blotting (WB, also known as immunoblotting or protein blotting) is a widely used laboratory method to detect specific protein characteristics, including abundance, apparent molecular mass, PTMs, and splice variants, in a sample of tissue homogenate or cellular extract (Jensen, 2012; Mahmood Yang, 2012).
