To obtain linear signals with the majority of western blots, we recommend loading smaller amounts of protein sample between 1 and 10 g per well. To avoid under- or overloading samples, determine the protein concentration of each sample prior to electrophoresis with a compatible protein assay.
In subject area: Nursing and Health Professions. Blotting refers to the transfer of macromolecules, such as proteins or nucleic acids, to a thin sheet of derivatized paper or adsorptive membrane matrix, allowing for easier and faster detection.
Protein extract should not be too diluted to avoid loss of protein and large volumes of samples to be loaded onto gels. The minimum recommended concentration is 0.1 mg/mL, optimal concentration is 15 mg/mL).
Consider the concentration of your purified protein, lysate, or culture sample. Ideally, it is best to load 2 g per well of a purified protein or 20 g of a complex mixture like whole cell lysates if you are doing Coomassie stain only.
It detects viral antigens (proteins usually on the surface of viruses) using antibodies against those proteins. A positive Western blot indicates the presence of viral antigen which very often means live virus in our patient. That patient may have an ongoing viral infection.
Dilute protein samples in buffer to final protein concentrations of 1100 ng/l. Tip: The protein of interest is diluted in dilution buffer for denaturing conditions, dilution buffer for native conditions, or another preferred buffer. Apply 1 l samples of diluted protein directly onto membrane.
Avivas Western Blot Conditions: Typical antibody concentration range = 0.2 - 5.0 ug/mL, or begin with a 1:1000 dilution of antibody reconstituted at 1 mg/mL.
