Take a look at this image. What might this be? A frightening monster? Two friendly bears? Or something else entirely? For nearly a century, ten inkblots like these have been used as what seems like an almost mystical personality test. Long kept confidential for psychologists and their patients, the mysterious images were said to draw out the workings of a persons mind. But what can inkblots really tell us, and how does this test work? Invented in the early 20th century by Swiss psychiatrist Hermann Rorschach, the Rorschach Test is actually less about the specific things we see, and more about our general approach to perception. As an amateur artist Hermann was fascinated by how visual perception varies from person to person. He carried this interest to medical school, where he learned all our senses are deeply connected. He studied how our process of perception doesnt just register sensory inputs, but transforms them. And when he started working at a mental hospital in eastern Switze

Problem: Uneven spots on blot or speckled background A speckled or spotty background can be caused by foreign material adhering to the membrane, antibody aggregation, or dirty scanner or cassette surfaces. All are preventable by fastidious handling procedures.
5 Steps to Reducing Background in Western Blots STEP 1: Use clean, fresh buffers. STEP 2: Use the correct blocking agent. STEP 3: Dont skimp on the wash steps! STEP 4: Find the best exposure time for your chosen detection method. STEP 5: Optimize your antibody concentrations.
To reduce high background due to high antibody concentration, titrate the antibodies. A dot blot can be used to titrate both primary and secondary antibodies in a checkerboard like pattern. Blocking the membrane is a crucial step that prevents non-specific binding of antibody to the membrane.
Wash that black right out of your blot Washing removes excess antibody that is not bound specifically to a protein. Wash your blot with at least 3 changes of wash buffer, incubating for 5 minutes at room temperature for each wash. Agitate the blot while washing and dont skimp on the amount of wash buffer you use.
A blotchy background can be due to a dry membrane or insufficient washing; both problems are readily solved by ensuring that the blot is immersed at all times. Additional handling precautions should be taken to prevent accidental contamination from common laboratory sources.
Background noises include environmental noises such as water waves, traffic noise, alarms, extraneous speech, bioacoustic noise from animals, and electrical noise from devices such as refrigerators, air conditioning, power supplies, and motors.
Western Blot possible causes solutions for high background Too much protein per lane. Titrate down the amount of protein loaded per lane. Insufficient blocking of non-specific binding. Adjust blocking conditions.
