A Southern blot is a laboratory method used to detect specific DNA molecules from among a many other DNA molecules.
Northern blotting is a molecular biology technique used to study gene expression by detecting and analyzing specific RNA molecules in a sample. It involves electrophoresis to separate RNA fragments by size and hybridization with a labeled probe to identify target RNA sequences.
Southern blotting \textbf{Southern blotting} Southern blotting is used as hybridization method that detects a specific target gene of interest among other DNA molecules present in a sample.
Southern blotting is used to detect specific DNA sequence in a given sample of DNA. The first step in a Southern blot is to prepare the DNA mixture by breaking it into small fragments using a restriction enzyme. The mixture of DNA fragments is then separated ing to size by gel electrophoresis.
The bands formed on the gel are then transferred to a nitrocellulose paper or nylon membrane through the technique called hybridization (blotting technique).
Southern blotting allows the detection of a given DNA sequence in a complex mixture of DNA sequences. It can be used to identify homologous sequences in genomic DNA, or to facilitate gene mapping through restriction mapping of genes or in the detection of restriction fragment length polymorphisms.
Western blotting techniques are used to transfer the protein molecules from acrylamide gels to membranes for further procedures. Explanation: Western blotting involves a transfer of electrophoresed protein bands from a polyacrylamide gel on to a nitrocellulose or nylon membrane.
Compared with chromatography, the Southern blot approach increases the detection resolution to a level that may be useful for examining specific genomic loci. However, it still requires a large amount of starting DNA, and the efficiency of restriction enzyme activity could affect the detection accuracy.
