In this video, I will show you how to perform a Western blot. The purpose of a Western blot is to specifically look at a single protein and visualize it on a camera. The steps included in running a Western blot are a protein gel, which can be found in a different video. But using the product of this protein gel, the proteins themselves are transferred onto a membrane. A primary antibody is added to the membrane to specifically bind to your protein of interest. A secondary antibody binds the first antibody, and the secondary antibody usually contains some sort of fluorophore, which we are then able to see when placing everything into a camera system. Blotting is a technique by which a macromolecule such as DNA, RNA, or protein is resolved in a gel matrix, transferred to a solid support, and detected with a specific probe. These powerful techniques allow the researcher to identify and characterize specific molecules in a complex mixture of related molecules.
Northern blot is a laboratory analysis method used to study RNA. Specifically, purified RNA fragments from a biological sample (such as blood or tissue) are separated by using an electric current to move them through a sieve-like gel or matrix, which allows smaller fragments to move faster than larger fragments.
In molecular biology and genetics, a blot is a method of transferring large biomolecules (proteins, DNA or RNA) onto a carrier, such as a membrane composed of nitrocellulose, polyvinylidene fluoride or nylon.
Southern blotting is useful not only for detecting the presence of a DNA sequence within a mixture of DNA molecules, but also for determining the size of a restriction fragment in a DNA sample.
The blotting methods are fairly simple and usually consist of four separate steps: electrophoretic separation of protein or of nucleic acid fragments in the sample; transfer to and immobilization on paper support; binding of analytical probe to target molecule on paper; and visualization of bound probe.
