[MUSIC PLAYING] NICO WAGNER: Hi, Iamp;#39;m Nico and Iamp;#39;m a PhD student in the Molecules, Cells, and Organisms program at Harvard University. In this video, I will show you how to perform a Western blot. The purpose of a Western blot is to specifically look at a single protein and visualize it on a camera. The steps included in running a Western blot are a protein gel, which can be found in a different video. But using the product of this protein gel, the proteins themselves are transferred onto a membrane. A primary antibody is added to the membrane to specifically bind to your protein of interest. A secondary antibody binds the first antibody, and the secondary antibody usually contains some sort of fluorophore, which we are then able to see when placing everything into a camera system. To run a Western blot, the first step after running the protein gel is to assemble the Western sandwich. The Western sandwich consists of three sheets of Whatman paper, the gel, a nitrocellulo

It detects viral antigens (proteins usually on the surface of viruses) using antibodies against those proteins. A positive Western blot indicates the presence of viral antigen which very often means live virus in our patient. That patient may have an ongoing viral infection.
Blotting is a technique by which a macromolecule such as DNA, RNA, or protein is resolved in a gel matrix, transferred to a solid support, and detected with a specific probe. These powerful techniques allow the researcher to identify and characterize specific molecules in a complex mixture of related molecules.
A western blot is a laboratory method used to detect specific protein molecules from among a mixture of proteins. This mixture can include all of the proteins associated with a particular tissue or cell type.
techniques are used to detect and analyze three types of biological macromolecules: DNA, RNA and proteins. Results of a blotting experiment tell you whether a macromolecule of a specific sequence is present in your sample or not.
The currently licensed Du Pont Western blot test specifies that the test result should be interpreted as positive only when the detected bands include p24 and p31, and gp41 or gp120/160 (12) (see Table 2).
In reading this western blot, note that each condition (i.e., cell line) is loaded in a column. The relative level of a given protein is evaluated across its respective row. For example, this gel shows that Cathepsin D is much more prominent in the Capan-2 cell line than the AsPC-1, BxPC-3, or Capan-1 cell lines.
A Western blot test is typically used to confirm a positive HIV diagnosis. During the test, a small sample of blood is taken and it is used to detect HIV antibodies, not the HIV virus itself.
