The dye molecules bind to proteins, including those in wool (keratin), to form a proteindye complex. As with all staining methods, Coomassie staining detects some proteins better than others, based on the chemistry of action and differences in protein composition. Thus, Coomassie staining can detect as little as 810 ng per band for some proteins and 25 ng per band for most proteins.

Principle Coomassie Blue R250 is an anionic dye that stoichiometrically binds to proteins, and is therefore preferable to silver staining methods for estimation of relative abundance of proteins useful for differential expression analysis of (2-DE) gels.
