What are the Similarities Between Rho Dependent and Rho Independent Termination? Rho dependent and Rho independent termination are two major termination strategies found in prokaryotes. Both forms release transcript from the DNA template to terminate the transcription.
Translation ends in a process called termination. Termination happens when a stop codon in the mRNA (UAA, UAG, or UGA) enters the A site. Stop codons are recognized by proteins called release factors, which fit neatly into the P site (though they arent tRNAs).
Transcription termination occurs when a transcribing RNA polymerase releases the DNA template and the nascent RNA. Termination is required for preventing the inappropriate transcription of downstream genes, and for recycling of the polymerase.
Rho-dependent termination occurs by binding of Rho to ribosome-free mRNA, C-rich sites being good candidates for binding. Rhos ATPase is activated by Rho-mRNA binding, and provides the energy for Rho translocation along the mRNA; translocation requires sliding of the message into the central hole of the hexamer.
Transcription termination is caused by the destabilization and/or a conformational change of the Pol II EC after transcribing the poly(A) site. Release of antitermination factors (left) or recruitment of termination factors (right) triggers dissociation from template DNA.
Two signals direct RNA polymerase II (pol II) to stop transcription: the previously identified 3 end formation signals located close to the poly(A) site and an additional downstream element (DSE) located at the region of termination. The downstream signal (135 bp) appears to act by pausing the elongating polymerase.
