Transcription factors are proteins that bind to DNA-regulatory sequences (enhancers and silencers), usually localized in the 5-upstream region of target genes, to modulate the rate of gene transcription.
Introduction. Transcription factors (TF) control gene expression by binding to their target DNA site to recruit, or block, the transcription machinery onto the promoter region of the gene of interest. Their function relies on the ability to find their target site quickly and selectively1.
How do enhancers affect transcription? Genome profiling has revealed that general transcription factors (GTFs) and RNA polymerase II (Pol II) are recruited to enhancers24. Thus, it appears that enhancers serve as centres for the assembly of the pre-initiation complex (PIC).
The DNA sequence that a transcription factor binds to is called a transcription factor-binding site or response element. Transcription factors interact with their binding sites using a combination of electrostatic (of which hydrogen bonds are a special case) and Van der Waals forces.
Transcription factors recognize and bind to specific DNA sequences called promotor or enhancer sequences. Promotor sequences are typically upstream of a gene and ensure accurate initiation of transcription.
TFs bind to enhancers in a combinatorial manner, which is facilitated through direct and indirect cooperative mechanisms. The combinatorial nature of enhancer occupancy allows genes to be regulated in complex patterns in both space and time.
