Pap test. During a Pap test, a tool called a speculum holds the vaginal walls apart. A sample of cells from the cervix is collected using a soft brush and a flat scraping device called a spatula (1 and 2). The cells are placed in a bottle that contains a solution to preserve them (3).
The principle of pap stain Solution 1 has Hematoxylin stain, a basic nuclear stain. Solution 2 has OG-6, an acidic cytoplasmic stain, has orange G with phosphotungstic acid (6 denotes the used concentration of phosphotungstic acid). Orange G is an acid dye and stains keratin with a bright intense orange.
Described pap staining liquid includes improvement haematoxylin dyeing liquid, EA50 dyeing liquor and orange G dyeing liquor. Wherein, improvement haematoxylin dyeing liquid includes following components: sodium iodate, HAEMATOXYLIN, aluminium potassium sulfate, ethanol, glacial acetic acid and water.
Kit Components: 0.5% Periodic Acid Aq (250 ml) Schiff Reagent (250 ml) 0.55% Potassium Metabisulfite (250 ml) Harris Hematoxylin Acidified (250 ml) Phosphate Citrate Buffer pH 5.0 (250 ml) Diastase Powder (0.5 gm)
Poly(A)-binding protein promotes export from the nucleus and translation, and inhibits degradation. This protein binds to the poly(A) tail prior to mRNA export from the nucleus and in yeast also recruits poly(A) nuclease, an enzyme that shortens the poly(A) tail and allows the export of the mRNA.
The Papanicolaou stain is undoubtedly the most commonly used in cytology. It has three cytoplasmic dyes, orange G, eosin Y, and light blue. It can stain cells blue or blue-green (basophilia, cyanophilia), pink (acidophilia, eosinophilia), orange (orangeophilia), or indeterminate (gray-blue).
