Lets say that single bacteria colony is suspended in 10 ml of broth. Then, 0.1 mL of this suspension is further diluted into 10 mL of broth. It is assumed that this diluted suspension has a count of 10^5 cfu/ml.
The 600 nm wavelength is specifically chosen for bacterial OD measurement because it is the known wavelength that is safe and not harmful to bacteria compared to UV wavelengths (5). Additionally, other wavelengths can scatter light when they encounter DNA, RNA, or proteins.
Expected OD 600 is determined by counting cell number using an alternative technique (for example microscope slide method) and converting to OD 600 using the rule of thumb that 1 OD 600 = 5 x 108 cells/ml for E. coli.
The fundamental principle behind OD600 is the scattering and absorption of light by bacterial cells, both live and dead, as well as any particles in the samle. The OD600 reading provides an indirect estimate of the cell density in a sample.
Measuring the OD600, or the optical density (OD) at 600 nm, is the easiest way to gauge the bacterial culture growth stage. Optical density measures the degree of light scattering caused by the bacteria within a culture; the more bacteria there are, the more the light is scattered.
We found an E. coli cell number of 2.66 x 10^9 cells per 1 OD600 unit, this is within the data found in the literature. The counting range is from 500 million to 6 billion E. coli cells per millilitre.
The 600-nm wavelength is specifically chosen for bacterial OD measurements because unlike UV wavelengths, 600 nm is not harmful to the culture. This wavelength is also not usually absorbed by yellow-ish media like TSB (tryptic soy broth) and LB (lysogeny broth).
APHA method (APHA, 2012) indicates 750, 664, 647 and 630 nm as the wavelengths most appropriate for measuring O.D. in microalgae cell density calculations. From these we selected 750 nm for all species as it is a wave length well beyond the absorbance peak of the chlorophylls in all species examined.
