The Papanicolaou stain is undoubtedly the most commonly used in cytology. It has three cytoplasmic dyes, orange G, eosin Y, and light blue. It can stain cells blue or blue-green (basophilia, cyanophilia), pink (acidophilia, eosinophilia), orange (orangeophilia), or indeterminate (gray-blue). Ethyl alcohol is the fixative specifically recommended for cytological preparations. Fixation coarsens the cell structures and sharpens nuclear chromatin pattern and its details.
Pap Staining involves five dyes in three solutions, a nuclear stain and two counter stains: Nuclear Stain: Harris Haematoxylin which is used to stain cell nuclei. Counter Stain OG6: Orange G dye is used to stain keratin. Its original role was to stain the small cells of keratinizing squamous cells present in sputum.
Saline solution may be used for moistening. The lubricant does not dissolve during processing of the slide. It often stains dark purple and may hide numerous cells. Before scraping, the wooden spatula should be dipped in saline to moisten it and prevent too rapid dehydration of the scraped cells.
Wash slides in deionized or distilled water for 5 min at room temperature. Destain slides in 0.5% HCl in 70% ethanol for 5-15 min at room temperature.
