Several factors may affect the results of Gram staining: If the smear is too thick, proper decolorizing will not be possible. If the smear is overheated during heat fixing, the cell walls will rupture. Concentration and freshness of reagents may affect the quality of the stain.

Crystal violet is the primary stain that is used to stain the bacterial smear on the slide. Iodine, which is a mordant, supports the adherence of the violet color to the cell wall.
Gram staining is a common technique used to differentiate two large groups of bacteria based on their different cell wall constituents. The Gram stain procedure distinguishes between Gram positive and Gram negative groups by colouring these cells red or violet.
Answer and Explanation: Normally, Gram-positive organisms appear purple using the Gram stain. However, if you forgot to use crystal violet, they would take up the counterstain (safranin) and appear pink.
The gram stain utilizes crystal violet as the primary stain. This basic dye is positively charged and, therefore, adheres to the cell membranes of both gram negative and positive cells. After applying crystal violet and waiting 60 seconds the excess stain is rinsed off with water. Next, a mordant is used.
Without the crystal violet staining, it would be impossible to differentiate between Gram-positive and Gram-negative bacteria, as the secondary counterstain, usually safranin, would stain all cells pink instead of just the decolorized Gram-negative cells.
A safranin counterstain is used to stain these Gram-negative cells pink. However, if the safranin counterstain were forgotten, the Gram-negative bacteria would remain unstained, as the original crystal violet stain would have been removed during the ethanol wash, and no additional stain would have been applied.
If there is a reversal of crystal violet and safranin stains, then the cross-link between iodine and crystal violet wont happen, and thus during decolorization the safranin will fade away, leaving all of the cells colorless.
