Special stains are processes that generally employ a dye or chemical that has an affinity for the particular tissue component that is to be demonstrated. They allow the presence/or absence of certain cell types, structures and/or microorganisms to be viewed microscopically.
Basic Procedure for Papanicolaou Staining (Pap stain) Fix the smear with 95% Ethanol 15 minutes. Rinse in tap water. Add the Harris Hematoxylin dye for 1-3 minutes. Rinse in tap water or Scotts tap water. Dip the preparation in 95% Ethanol 10 dips. Add orange G-6 stain for 1.5 minutes. Dip in 95% Ethanol 10 dips.
Staining techniques The Papanicolaou stain (PAP) requires immediate fixation in alcohol. Nuclear chromatin patterns are easily discerned and the degree of maturation/keratinisation of squamous cells is readily appreciated. Mucin may not be seen so easily. May-Grnwald Giemsa (MGG) staining is used on air-dried smears.
If theres wax remaining in the tissue section, the staining solution cant penetrate the specimen properly, which leads to weak staining. Return slide to fresh xylene to remove paraffin wax from section then proceed with procedure to re-stain tissue specimen.
Pap Staining involves five dyes in three solutions, a nuclear stain and two counter stains: Nuclear Stain: Harris Haematoxylin which is used to stain cell nuclei. Counter Stain OG6: Orange G dye is used to stain keratin. Its original role was to stain the small cells of keratinizing squamous cells present in sputum.
Therefore, color/stain normalization is an important process that involves adjusting the mean color of one image to another image, with the aim of minimizing color variations between them.
Papanicolaou stain (also Papanicolaous stain and Pap stain) is a multichromatic (multicolored) cytological staining technique. The Papanicolaou stain is one of the most widely used stains in cytology, where it is used to aid pathologists in making a diagnosis.
The conventional Pap stain has 24 solutions, of which three are acid dyes, six water, one is chemically defined bluing agent, eight 95% ethanol, three absolute alcohol and three xylene Gill, 2006.
