Staining is done to visualise the specimen clearly under the microscope. Without staining the specimen under the microscope will look transparent or will not be visible clearly. Hence, staining is done to make the components of cell clearly visible. For example, saffranin is used to stain the nucleus.
For example, one of the most common stains, Hematoxylin, is a basic dye that stains proteins a blue color, while Eosin stains proteins a pink color. These two stains are commonly used together to define intracellular organelles and proteins.
Hematoxylin shows the ribosomes, chromatin (genetic material) within the nucleus, and other structures as a deep blue-purple color. Eosin shows the cytoplasm, collagen, docHub tissue, and other structures that surround and support the cell as an orange-pink-red color.
Numerous forms of special stains have been used to detect collagen fibers but the picrosirius red stain demonstrated superior results compared to the other special stains. The collagen fibers were stained intensely with spectacular birefringence.
Periodic Acid Schiff (PAS) This stains cells that are rich in carbohydrates. Positive cells are macrophages, docHub tissue (collagen and basement membranes), and fungi. The benefit of staining a section with PAS is that the background is also stained and can be used for confirmation purposes.
Macenko stain normalization technique The color transformation to OD values provides a space where a linear stain fusion yields a linear fusion of OD values. Subsequently, the transparent pixels are removed if the OD value is below a specific threshold. The OD value is split into two matrices, given by Eqs.
Diagnosis of Collagen Type III Glomerulopathy Using Picrosirius Red and PASH/Massons Trichrome Stain.
Masons Trichrome stain is used to highlight docHub tissue fibers in a tissue section. Collagen fibers are stained a blue hue, nuclei a blue-black, and cytoplasm, keratin, and muscle fibers a range of pink to red hues.
