Tissue embedding is the process of placing tissues in molten paraffin before sectioning, using methods like metal molds or pans, to facilitate rapid embedding and ensure proper orientation for accurate morphology demonstration in samples.
A protein defined as amyloid must satisfy two basic criteria: (1) the protein is present in an insoluble fibrillar form that deposits in tissues, ultimately associated with a disease condition; and (2) the deposited proteins are stained by Congo red with concomitant apple-green birefringence under cross-polarized light
Biopsy. A tissue sample can be checked for signs of amyloidosis. The biopsy may be taken from the fat under the skin on the abdomen or from bone marrow. Some people may need a biopsy of an affected organ, such as the liver or kidney.
A common laboratory method that uses two dyes called hematoxylin and eosin that make it easier to see different parts of the cell under a microscope. Hematoxylin shows the ribosomes, chromatin (genetic material) within the nucleus, and other structures as a deep blue-purple color.
Most commonly, amyloid can be identified with the Congo red stain as an apple-green birefringence when viewed under polarized microscopic light. Additionally, other stains may be used to identify amyloid such as crystal violet or thioflavin T.
The HE staining method involves application of haematoxylin mixed with a metallic salt, or mordant, often followed by a rinse in a weak acid solution to remove excess staining (differentiation), followed by bluing in mildly alkaline water.
Congo Red and Thioflavin S are the two major histological stains used to detect any form of amyloid. These dyes bind to the characteristic -pleated sheet conformation of amyloid.
Amyloid Identification Criteria It is weakly basophil with alcian blue and toluidine blue. It stains metachromatically with crystal violet and toluidine blue. It stains with congo red and sirius red F3B. Congo red and sirius red stained amyloid show green birefringence.
