Western blot is a technique to detect and analyze the protein of Interest it is based on the principle of antigen antibody interaction in the first step the gel is ran where the protein is basically separated on base of its molecular weight so among these protein present in the gel we need to find whether our protein of interest is present or not and that is detected using an antibody but first the content of the gel that means all the proteins are basically transferred into a pvdf membrane with Electro blotting technique then the membrane is incubated with primary antibody blocking solution and then the secondary antibody letamp;#39;s say the protein of interest is present so the primary antibody would detect that protein of Interest secondary board antibody would detect the primary antibody and a color reaction would be developed there are different modes of development like enzymatic chemiluminescent and fluorescent this is how a typical gel look like and there are wide variety of

What is a western blot? The term blotting refers to the transfer of biological samples from a gel to a membrane and their subsequent detection on the surface of the membrane.
I recommend always to use the facing side of the membrane (PVDF or nitrocellulose) to probe your antibodies, since in this side of the membrane your proteins are fixed and oriented.
The polyvinylidene fluoride (PVDF) or nitrocellulose (NC) membrane should always be oriented on the anode (+) side of the gel. Western blotting: Troubleshooting Guide for Protein Transfer Cytiva news-center 5-to Cytiva news-center 5-to
Put two pre-soaked blotting pads in the bottom of the electrode, then the first gel sandwich, with the MEMBRANE SIDE UP.
The pore size of the membrane, as well as the transfer buffer and the type and percentage of acrylamide gel used all affect protein transfer from a gel to a membrane. The necessity of and strategies for improving confidence in the - NCBI NCBI articles PMC4791038 NCBI articles PMC4791038
Unsupported nitrocellulose is innately fragile and is not recommended for stripping and reprobing. Supported nitrocellulose is an inert support structure with nitrocellulose applied to it. Edge Effect: When multiple lanes are run on a blot, the outer lanes may experience the edge effect. The edge effect occurs when proteins drift towards the center; luckily, this can be removed through the use of a loading control, fine-tuning the entire blotting process by correcting the variation in binding.
Since nitrocellulose membrane does not have to be activated like PVDF it only needs to be wetted using cold transfer buffer. How do you soak your nitrocellulose membrane before transferring ResearchGate post Howdoyousoak ResearchGate post Howdoyousoak
