Western Blot possible causes solutions for high background Too much protein per lane. Titrate down the amount of protein loaded per lane. Insufficient blocking of non-specific binding. Adjust blocking conditions.
The more essential improvements needed for improving confidence in the accuracy of western blots include better documentation of primary antibodies, better training of researchers to use quantification software, use of antibodies within their linear range and proper choice of a loading control.
If the Western blot shows high background signal, revising the washing steps on the next run-through has proven to be useful. Increasing washing times and volumes will reduce background signal along with performing additional buffer changes or using a stronger detergent (e.g., SDS instead of Tween 20).
5 Steps to Reducing Background in Western Blots STEP 1: Use clean, fresh buffers. STEP 2: Use the correct blocking agent. STEP 3: Dont skimp on the wash steps! STEP 4: Find the best exposure time for your chosen detection method. STEP 5: Optimize your antibody concentrations.
If you have run the gel at a high voltage for a short period of time, the proteins may not run in a uniform manner, causing blurry bands. Blurry bands can also occur if a mistake is made during the preparation of the running buffer, or if bubbles were trapped during the protein transfer stages.
Blocking is critical for clean blots. Blocking agents such as non-fat dry milk or BSA occupy non-specific binding sites resulting in lower background. Check to see if there is a preferred blocking agent and concentration for your antibody. A 1-5% blocking solution is usually recommended.
Patchy and uneven spots on the blot are usually caused by improper transfer. If there are air bubbles trapped between the gel and the membrane, it will appear darker on the film. It is also important to use a shaker for all incubation, so that there is no uneven agitation during the incubation.
Blotchy, Flecked, Or Dirty Background These artifacts are most commonly the result of uneven coating of buffer or antibody, the membrane drying out, or aggregates forming in the antibody or blocking buffer. Follow the tips below to identify and solve the cause of your imperfect western blot background.
