Dot blot is one of the easiest ways to determine whether a protein of interest is present in a sample. It is also useful for checking antibody specificity, and for optimizing antibody titers prior to running a Western blot.
Dot blot is a technique for detecting, analyzing, and identifying proteins. This technique is similar to western blot, but protein samples are not separated using electrophoresis; instead, proteins are spotted through circular templates directly onto the membrane or paper substrate.
There are two built in methods for analyzing a dot blot in ImageJ. The first is to treat each row as a horizontal lane and use ImageJs gel analysis function. The second is to subtract the background and measure the integrated density of each dot. Figure 1.
Adjust pH to 8.0 with NaOH. Dilute protein samples in buffer to final protein concentrations of 1100 ng/l. Apply 1 l samples of diluted protein directly onto membrane. After applying the samples, the membrane should be dried for a short time at room temperature before proceeding with the detection process.
During a dot blot, the sample (typically a cell or tissue lysate, or a recombinant protein) is spotted directly onto a nitrocellulose or PVDF membrane, which is then blocked prior to incubation with the primary antibody.
