FLAG tags are epitope proteins with DYKDDDDK sequence, where D=Aspartic acid; K=Lysine; Y=Tyrosine. Its a highly specific protein used for assays that involve antibodies for identification, like protein purification and immunological assays, such as immunoprecipitation and immunofluorescence.
FLAG Tag Usage in Recombinant Protein Production Adding a FLAG-tag to the N- or C-terminus of a protein allows rapid purification of the over-expressed recombinant protein, by using a FLAG-tag specific monoclonal antibody conjugated to agarose beads.
FLAG is an octapeptide epitope tag with the amino acid sequence DYKDDDDK that can be added to the N-terminus, C-terminus, or within a recombinant protein. Note that some antibodies may only recognize the FLAG epitope in certain positions (such as N- or C-terminus only).
It is one of the most specific tags and it is an artificial antigen to which specific, high affinity monoclonal antibodies have been developed and hence can be used for protein purification by affinity chromatography and also can be used for locating proteins within living cells.
Anti-FLAG antibodies are reagents that can be used to detect or stain Flag-tagged recombinant proteins. The FLAG epitope tag, is an octapeptide containing the amino acid sequence DYKDDDDK.
The M2 anti-FLAG antibody binds N-terminal FLAG, C-terminal FLAG, and Met-FLAG fusion proteins. The M1 anti-FLAG antibody binding is calcium-dependent and specific for the N-terminus of the FLAG tag.
