Dot blot is a method for detecting biological samples like proteins or nucleic acids, similar to western blotting and southern blotting but samples are dotted onto a membrane instead of being blotted from a gel. It is used for applications such as monitoring labeling efficiency, estimating protein concentration, and comparing antibody performance. To determine labeling efficiency with biotin, compare the labeled sample to a biotinylated reference standard by making serial dilutions of both samples in the same way. A 10-fold serial dilution is a good starting point for a wide range of samples.

Dot blotting is a simple, convenient method for detection of proteins in crude lysates or solutions without the need for separation by SDS-PAGE. This method is especially useful as a simple control because it avoids problems that may be due to the western transfer process.
In conclusion, the dot-blotting technique described in this report provides an accurate, more efficient, and possibly more reliable method for the assessment of in vivo proliferation compared with conventional immunohistochemical determination of BrdU-labeling index.
Loading too much protein leads to signal saturation in western blots, yet too little produces weak signals. This protocol describes an assay development experiment to determine the appropriate protein load for target and control detection prior to performing the actual western blot experiment.
Therefore, nitrocellulose could not be used for electroblotting because high ionic strength buffers are needed for efficient retention of DNA [34]. PVDF was investigated as an alternative because it has been used in capillary transfer with low ionic strength buffers and binds both dsDNA and ssDNA [39, 40].
Before staining, moisten the PVDF membrane in methanol for several seconds, wash in double-distilled, water, then soak with transfer buffer for 3 minutes. Alternatively, re-wet the membrane by incubating it in buffered saline solution* containing 0.5% Tween 20* (v/v) for 15 to 30 minutes.
Dot-blot is semi-quantitative method to estimate the presence of specific DNA sequences based on hybridization. DNA samples are spotted next to each other on a filter (nitrocellulose or nylon) in dots of uniform diameter. The filter is subject to denaturing conditions and then hybridized with a probe.
A speckled or spotty background can be caused by foreign material adhering to the membrane, antibody aggregation, or dirty scanner or cassette surfaces. All are preventable by fastidious handling procedures.
The size of the target protein should be considered when choosing transfer conditions. Smaller proteins will transfer out of the gel faster and may actually transfer through the PVDF membrane into the filter papers beyond. PVDF with a smaller pore size can be used for small proteins and peptides.
Dot and slot blotting are simple techniques for immobilizing bulk unfractionated DNA on a nitrocellulose or nylon membrane. Hybridization analysis can then be carried out to determine the relative abundance of target sequences in the blotted DNA preparations.
Sample Preparation Wash cells twice to remove residual media using PBS. Remove PBS and add appropriate volume of RIPA Buffer plus protease inhibitors (1 ml per 0.5 to 5 x 107 cells). Incubate at 4C for 5 min. Spin the lysate at 8000 x g for 10 min at 4C to pellet the cell debris.
