Schmorls method uses the reducing properties of melanin to stain granules blue-green. The most specific method of all is an enzyme histochemical method called DOPA-oxidase. It requires frozen sections for best results, but paraffin sections of well-fixed tissues may be used.
Iodine fixes the crystal violet into the cell call of bacteria by acting as a chemical mordant. G+ bacteria remain purple due to their adherence to the iodine. Iodine binds to starch, making a blue-black color. This gives color to the epidermal cells, making them more readily visible and observable.
Amylose in starch is responsible for the formation of a deep blue color in the presence of iodine. The iodine molecule slips inside of the amylose coil. Iodine - KI Reagent: Iodine is not very soluble in water, therefore the iodine reagent is made by dissolving iodine in water in the presence of potassium iodide.
Iodine staining is a coloring reaction that occurs by complexing iodine with crystalline compounds or polymers. The molecular origin of the color is the formation and subsequent alignment and orientation of polyiodide ions.
Basic stains, having a positive charge, bind strongly to negatively charged cell components such as bacterial nucleic acids and cell walls.
The binding proteins include thyroxine binding globulin (TBG), thyroxine binding prealbumin (TBPA) and thyroxine binding albumin (TBA). Binding of hormones apart from serving as a reservoir also helps to prevent urinary loss of hormones.
The HE stain provides a comprehensive picture of the microanatomy of organs and tissues. Hematoxylin precisely stains nuclear components, including heterochromatin and nucleoli, while eosin stains cytoplasmic components including collagen and elastic fibers, muscle fibers and red blood cells.
